Özet (EN)
HSV is one of the most important causes of corneal disease and loss of vision. Despite of high prevalance, mechanisms of reccurence and blinding complications are not obvious. Early diagnosis and treatment is important in the control of vision threatening complications. Clinical improvement can be achieved by antiviral treatment but they are not effective in the latent Herpes infections. In severe cases AMT is an alternative treatment choise. The aim of this study was to evaluate the effect of acyclovir and human amniotic membrane on herpes virus culture by analysis of viral DNA with RT-PCR.This study was conducted in Ankara University Veterinary Faculty Virology Department laboratory. MDBK cell culture and BHV-1 types were used. Out of two groups the first culture group was inoculated by absorbted virus and then amniotic membrane, acyclovir or both were added. In the second group virus inoculation wasn?t done, only applications similar to cell cuture were performed. After mebrane and drug application, culture was evaluated on 24th and 48th hours for CPE positivity with tissue culture microscope. In CPE (+) samples DNA was extracted for viral DNA analysis by real time PCR.After analysis Ct was found as VZKeyword: Amnion = Amnion ; Asiklovir = Acyclovir ; DNA = DNA ; Herpes simplex = Herpes simplex ; Hücre kültürü = Cell culture ; Keratit = Keratitis ; Polimeraz zincirleme reaksiyonu = Polymerase chain reaction ; Virüsler = Viruses
