Özet (EN)

Tomato Spotted Wilt Virus (TSWV) is one of the important plant virus diseases which limitates the pepper production in our country and the world. In this study, the development of the new marker(s) has been aimed in order to remove the disadvantages of the present molecular testing method which is used in breeding works to improve a type resistant to TSWV. Totally 96 pepper plants on F4 rank have been used as study materials. By using SCAR marker developed by Moury et. al (2000), related gene spot has been multiplied and genotypes' resistance to TSWV at codominant level (RR, Rr, rr) after two seperate CAPS phase has been determined. In the first part of the study to develop new markers, totally 58 primer combinations have been tried by generating SCAC568 primer combinations with SRAP primers. In the second part of the study, 29 SSR primers most of which are mapped, and developed for pepper, have been used. The primers determined as polymorphic with Bulk Segregation Analysis method has been evaluated as marker candidates. Stating that the results obtained from Hpmse031 SSR primer show consistency with the results from the present molecular testing of TSWV, PCR amplification products of the related primer have been viewed by using agarose gel and Fragment Analyzer system. Apart from 96 pepper plants on F4 rank, the trial of the marker has been practiced on 381 pepper genotypes at different genetic substructure. The marker newly developed at molecular testing of the resistance to TSWV will provide advantages in terms of work force, time and finance as it removes enzyme fraction steps.
