Investigation of the possibilities for reproduction of grape varieties by shoot tip culture method in Bilecik irikarasi, Sarı Üzüm, Kartal Çavuş and Razakı grape varieties that produced in Bilecik

In this study, it was aimed to investigate the reproduction possibilities of the local grape varieties of Bilecik İrikarası, Sarı Üzüm, Kartal Çavuş and Razakı grown in Bilecik by shoot tip culture, which is one of the tissue culture techniques, for clonal propagation. In addition, the propagation possibilities of local varieties that were not reproduced by in vitro shoot tip culture method in earlier studies were determined by using this method. MS (Murashige and Skoog, 1962) was used as nutrient medium in the study. 1 mg/l BAP (Benzyl Amino purine) was added to the MS to promote shoot growth and 2 mg/l IBA (Indol-3-butyric acid) was added to promote root growth. As a result of the study, the rooting rate, root number, root length, shoot number, shoot length, leaf number and node number values were determined from Bilecik İrikarası, Kartal Çavuş, Sarı Üzüm and Razakı grape varieties. When the cultivars were compared in terms of rooting characteristics, the highest rooting rate was 54.7%, the highest root number was 3.71 per plant, and the highest average root length value was 17.93 cm from Bilecik İrikarası. When the cultivars were evaluated in terms of shoot length, the highest shoot length value was determined in Razaki cultivar with 5.72 cm. Similarly, considering the number of leaves and nodes in the shoots were determined in Razakı variety, with the highest leaf number value was 8.71, the highest node number value was 6.71. As a result of the study, local varieties that used as material in this study showed positive results in in vitro propagation. However, it was determined that the concentration of IBA (Indol-3-butyric) and BAP (Benzyl Amino purine) to be added to the MS medium can be needs to be determine according to the varieties.
