Isoprenoids are found in many places in nature and range from basic cell components to secondary metabolites. Natural products such as terpenoids (isoprenoids) are usually obtained by extracting from their original sources, because the commercial synthesis of such complex structures, which often involve more than one chiral center, is very difficult and uneconomical. The isoprenoid biosynthetic pathway has attracted much attention in terms of metabolism engineering and significant progress has been made in increasing the metabolic flow on these pathways. A good blinen isoprenoid, taxol is a potent anticancer drug widely used in the treatment of various cancers. The original source of taxol is Taxus brevifolia and the amount obtained from the bark of this tree is quite low. In this study, I tried to increase the amount of taxol pioneers in Saccharomyces cerevisiae by using a model organism via metabolism engineering strategies. Taxol precursors are produced in the mevalonate pathway and the mevalonate pathway is fed by acetyl-CoA pathway in the cytosol. Therefore, by changing the pHXT7, which is an inducible promoter to direct the flow to acetyl-CoA, we have examined the change in the different mutant strains by changing GC-MS analysis.
