Effect of lycopene on apoptosis and testicular histology in bisphenol A is applied to Wistar Albino rats

Özet (EN)

Objective: Bisphenol A (BPA), food and beverage packaging monomer of polycarbonate plastic and epoxy resin is used as raw materials in which materials are endocrine disruptive chemicals. Lycopene belongs to carotene family is found naturally in fruits and vegetables is an effective antioxidant compounds resulting in the elimination of free radicals oxidative stress coming into the open, depending on various factors. According to the studies of BPA it has been demonstrated to have an adverse effect on many organs. In this study of BPA effects on the male testes histologically to reveal the genital organs and the possible effects of lycopene by reducing and / or eliminating we intend that. Materials and Methods: Dokuz Eylul University Faculty of Medicine Research Unit obtained from the Laboratory of Experimental Animals, Wistar albino strain total (180-200 g) were used 35 male rats. Wistar albino rats; 1. Control group (14 days no substance is not implemented) 2. Sham Group (0,1 ml of ethanol with 0.1 ml / 100g corn oil by oral gavage method for 14 days,) 3 Bisphenol Group (10 mg / kg of bisphenol A by oral gavage method during 14 days) 4. Lycopene Group (4 mg / kg of lycopene by oral gavage for 14 days) 5. Bisphenol A + Lycopene Group (10 mg / kg of bisphenol A and 4 mg / kg of lycopene by oral gavage method during 14 days), they were divided into 5 groups. All animal experiments are finished 12/12 hour dark / light cycle at 20-22 ° C, 50-60% relative humidity at room temperature which standard animal rooms and cages, aged tap water and were fed standard chow. Working with testicular tissue in assessing stereological methods (IHSS), the diameter of the seminiferous tubules were measured. Measurement results for each group of seminiferous tubule structure differences between the mean values, it groups the diameter of the tubules were compared. Hematoxylin-eosin-stained sections were evaluated by Johnsen's scores. All SPSS 21 statistical software in the mean ± standard error calculated on the difference between the groups after the Kruskal-Wallis, One-Way ANOVA, POST HOC were performed using LSD test and Student's t-test, p <0.05 significance level were taken into account. Results: According to the histologic and morphologic assessment made in the testicles, BPA Control group, Sham group and is corrupt according to the LKP and the LKP group LKP+ BPA was observed to prevent this disorder. The evaluation of DNA damage Control, Sham, LKP and the BPA + LKP group between spermatogoniy and spermatogenesis in significant differences were observed with TUNEL-positive staining cells and spermatogenesis in the BPA group spermatogoniy TUNEL positive cells stained with Control and Sham group was significantly higher than the LKP. Compared to BPA and BPA + LKP, BPA + LKP group in the TUNEL the number of positively stained cells showed a significant decrease compared to the BPA group. We did it with the purpose of supporting the active caspase result of TUNEL immunohistochemical (IHC), according to assessment; Control, Sham, LKP and BPA + LKP in spermatogoniyum between groups of active caspase IHC cells stained positively significant difference observed, in spermatogoniyum BPA group active caspase IHC positively stained cells as control, were higher in anlamlıderece according to the Sham and the LKP group. BPA and BPA + LKP group when the number of active caspase across the group IHC positively stained cells was observed a significant decrease compared to the BPA group. Between groups for MDA, GSH and GPX values compared; the experimental group is significantly higher than other groups while the MDA values of BPA, BPA group, GSH and GPX values were statistically significantly lower than the values of the other experimental groups. Also it is taken into consideration at the beginning of the end of the experiment by measuring the body weight of the subjects and experiments. Compared to all the groups in the pre-test and post-test subjects it showed increases in body weight and body weight of rats showed a statistically significant difference. Conclusions: : Our findings are in line with low-dose BPA's toxic damage caused by testicular 4mg / kg is LKP the protective effect, but we concluded that the damage is completely blocking.
