Abstract (EN)

This thesis has been performed in order to prepare molecule imprinted polymers (MIP) based on the metal-chelate interaction for the isolation and purification of the biomolecules (proteins, aminoacids, cholestrole, etc.) from their complex matrixes (blood, body fluids, etc.). For this purpose, firstly, a new generation bio-adsorbent having L-histidine memory in order to extract the proteins including surface histidine and differentiate D-L-histidine enantiomers has been devoloped and its chromatographic application has also been investigated. In the second step, a novel molecule imprinted adsorbent has been synthesized for the separation of 8-Hydroxy-2?-deoxyguanasine (8-OHdG) found in the different body fluids and used in the diagnosis of the oxidative DNA damage from its matrix and the availability of the obtained adsorbent in the solid phase extraction of 8-OHdG has also been investigated. In the last section, a novel chromatographic column material has been evolved for the determination of cholesterol which is a very important biomolecule for the human and efficiency of this column has been investigated. This study presents the first application for the preparative protein seperation using Fast Protein Liquid Chromatography (FPLC) and first attempt towards the imprinting of 8-OHdG in the literature. Moreover, all the MIPs that have been prepared within thesis have high selectivity towards related molecules, have low cost and are compatible with biological systems.
