Abstract (EN)
Artificial blood is a substance that carries oxygen, is compatible with all blood groups and can be purified from microorganisms. For this purpose, P(HEMA-co MACys) nanospheres were synthesized using MACys monomer in the first stage of the study. Fe3+-Protoporphyrin IX was covalently bound to these nanospheres based on the thio-ene reaction. In order for the synthesized P(HEMA-co-MACys-Fe3+-Protoporphyrin IX) nanospheres to bind and release oxygen reversibly, these nanospheres were transformed into P(HEMA-co-MACys-Fe2+-Protoporphyrin IX) to form a complex with serum albumin. Carbonic anhydrase (CA) is a metalloenzyme that converts carbon dioxide into bicarbonate and proton. In the second step of the study, a new cryogel-based chromatographic support material, PHEMA-pABSA, was synthesized for the purification of this enzyme. CA enzyme was purified and characterized using the PHEMA-pABSA cryogel column. Poly-ε-caprolactone (PCL) is a non-toxic biodegradable synthetic polymer with high permeability to many molecules. In the third stage of the study, this polymer was functionalized with chlorobis(2-2'-bipridyl)MATyr-Ru(II), a photosensitive amino acid monomer, and P(CL-Ru(II)bis(2-2'-bipyridyl)MATyr) was synthesized. Using this polymer as the encapsulating agent, ANADOLUCA concept, double emulsion solvent evaporation technique and EDC/NHS method, a new oxygen carrier system P(CL Ru(II)bis(2-2′-bipyridyl)MATyr-co-Hb-CA)-Fe2+/3+-Protoporphyrin IX nanospheres were synthesized and characterized. Leeches are bloodsucking creatures and their use in traditional medicine is increasing day by day. In the last phase of the study, the use of bionanoconjugates synthesized for artificial blood purposes by Hirudo verbana, a medicinal leech type, and its effect on hemoglobin types (oxyHb, metHb, deoxyHb) were investigated in detail.
