Abstract (EN)

In this study, biotinylation of primers from 5' end using cryogel column synthesized by ruthenium-based photosensitive cross-linking technique has been realized. For this purpose, firstly, DNA ligase cross-linked HEMA and monolithic photosensitive monomer based cryogel colum have been developed and characterized. In the second part of the study, avidity efficiency has been formed via binding of biotin to 5' end of the primer having Crimean Congo Hemorrhagic Fever (CCHF) by using synthesized enzyme based cryogel column system. The proof of this interaction has been performed by observation of increase the absorption of the samples at 354 nm using microplate reader. The efficiency of the designed primers having CCHF has been proved by using polymerase chain reaction (PCR). In the last step of this study, streptavidin cross-linked and photostable Quantum nanocrystals (QDs) having fluorescence property have been formed and biotinylated 5'-ssDNA probes have bound to streptavidin-QD platforms. Then, the fluorescence changes based on the hybridization of 3'-ssDNA complements which cause diagnosis of disease have been investigated.
