Investigations of in vitro antioxidant, wound healing and anticancer activities in U2OS osteosarcoma cells of Camellia sinensis extracts

Abstract (EN)

Camellia sinensis (L.) Kuntze (Theaceae) (tea) is a plant which is rich in polyphenols including flavan-3-ol (catechin) ve flavonol glycosides. It?s been reported that green tea which is obtained from C. sinensis has a potent antioxidant activity and also antiinflammatory, antimutagenic, anticarcinogenic, antiangiogenic, apoptotic, hipolipidemic, antiarteriosclerotic, antidiabetic, antibacterial, antiviral and antiaging activities. In this study, methanol and infusion extracts were prepared from the leaves of Camellia sinensis that were gathered from Trabzon Sürmene and the green tea that we bought in order to investigate antioxidant, wound healing and anticancer activities in vitro. Total phenol constituents were determined by Folin-Ciocaltaeu Method and DPPH (2.2-difenil-1-pikrilhidrazil) and ß-Caroten-Linoleik Acid System were used to determine antioxidant activity.The prepared C. sinensis extracts were given to NIH 3T3 cells at the concentrations of 0.5, 1, 2.5, 5, 10 ?g/ml and they were incubated for 24, 48 hours to detect wound healing effects by using Masson?s Trichrome Tecnique. The concentrations of extract (5, 10, 20, 40, 60 ?g/ml) were given U2OS osteosarcoma cells to evaluate their anticancer activities at 24, 48 hours by using MTT and flow cytometry assays.Total phenolic constituents and antioxidant activities of methonol extracts were higher than water extracts. The wound healing and anticancer activities of methanol extracts were significantly increased when compared to control. In the low concentrations of methanol extract groups in the wound healing effect, the observed numbers of mitotic cells were significantly high along with the total cell number. The 0.5 ?g/ml concentration of the methanol extract was indicated an increment in polygonal cell numbers when compared to control. There was no significance in the groups in respect to number of granule cells. In cytotoxicity test, it was observed that as the concentrations and incubation time increases the cytotoxic effect was also increased; whereas the proliferation was decreased. The apoptotic effect in methanol extracts were more potent when compared to infusion groups. Especially; the cells which were treated with 60 ?g/ml concentration of methanol extract were indicated higher apoptotic cell ratio.As a result it was concluded that methanol extracts of Camellia sinensis which have high total phenolic constituents were more effective in antioxidant, wound healing and anticancer activities
