Development of liquid chromatography mass spectrometry method for the determination of luteolin-7-glucoside and rutin in Campanula propinqua extracts

Abstract (EN)

In this study, a new LC-MS/MS method was developed for the determination of Rutin (RU) and Luteolin-7-glycoside (Lut-7G). During the studies, Zorbax C18 with an inner diameter of 3,5 μm was used. Optimum conditions were 100.0 V voltage, 30°C column temperature and used a mobile phase A, 0,5 % formic acid and B water:acetonitrile:formic acid (50:50:0,5 v/v/v). The method was validated according to ICH guideline. Applied weighted regression. A good linearity were optained in the concentration range of 0,975-1950 ppb for the rutin and 0,66-1200 pbb for the lut-7G. LOQ and LOD of the method were found to be 2,12 ppb-0,64 ppb for the rutin and 0,44 ppb-0,13 ppb for the lut-7G. The precision and accuracy of the method were satisfying with intra and interday recovery values of %98-99,4 for the rutin and %98,6-100,4 for the lut-7G and RSD values less than %2. The proposed method was applied to the leaf and flower extract of Campanula Propinqua. 46,57 µg/mg ve 4,07 µg/mg of rutin were found in the leaf and flower extract of the plant. Lut-7G was only found in the flower extract of the plant 4,57 µg/mg.
