Research on identification of mycotoxigenic fungi and main mycotoxin potantial in various cereal flours

Abstract (EN)
In this study, determination of mycotoxigenic fungi in various cereal flours from markets at Eskişehir/Türkiye and their capability of production of main mycotoxin were purposed.For this aim, 25 different flour samples were investigated and 543 mold isolates were obtained. The molds were identified at genus and species level according to traditional identification literatures. Chemotaxonomic data that was obtained metabolite profiles of all of the isolates with thin layer chromatography (TLC) method were used as promoter of morphological identification results. Mycotoxigenic fungi were screened with cultural methods and determined 130 molds as mycotoxigenic. Aflatoxins (B1, B2, G1, G2) and ochratoxin A production properties were measured with high performance liquid chromatography (HPLC) system. Aflatoxin production properties were detected between 0.13-10438.00 ng/ml, ochratoxin A production properties were detected between 3.69-28440.00 ng/g. 32 representative mycotoxigenic isolates were identified as Aspergillus flavus, A. japonicus, A. tubingensis, A. niger, A. ochraceus, A. alliaceus and Penicillium viridicatum with sequencing of internal transcribe spacer (ITS) regions, beta-tubulin and calmodulin gen regions.All of the flour samples were investigated with HPLC system for detection of mycotoxins. In addition to mycotoxin contamination at different levels were detected in 24 flour samples, we were determined 3 flour samples including aflatoxin, 2 flour samples including ochratoxin A above maximum limit values of Turkish Food Codex.
