Abstract (EN)

CXXC5 is an estrogen responsive gene whose deregulations are implicated in a number of diseases including cancers. It is crucial to understand the mechanisms through which CXXC5 expression is regulated for the characterization its role in physiologies and pathophysiologies. To gain an insight in the CXXC5 gene expression, we wanted to identify its promoter. We hypothesized that the promoter of CXXC5 could reside in close proximity to the transcription start site of its main transcript variant. In this study, we wanted to identify the main transcript(s) of CXXC5 and its transcription start site(s). CXXC5 has 14 transcript variants all of which encode the same protein. We found here that CXXC5 Transcript Variant 2 is the variant that has the highest expression levels in both MCF7 and HL60 cells, thus, it is the main transcript variant. CXXC5 Transcript Variant 2 has a broad transcription initiation region and by investigating the ranges of this region, we identified a novel transcript variant of CXXC5 as well.
