Abstract (EN)
The aim of this study is to investigate the Effects of Biocompatible Carriers Containing Vitamin D and PRP on Bone Defects Created in Rat Calvaria. We planned to investigate histopathologically, radiologically and biochemically with comparison. In the study, 52 male Sprague Dawley rats were used, except for the 4 used to obtain PRP(Platelet Rich Plasma) 5 mm bone defect was created and rats were randomly divided into 6 groups. Control group, chitosan group, chitosan+PRP group, PRP group, calcitriol+chitosan group, calcitriol+chitosan+PRP group. Calvarial defects were examined by sacrificing the rats in equal numbers at the 1st, 2nd, 4th and 8th weeks. The newly formed bone was evaluated histopathologically, microtomographically, macroscopically the blood contents analyzed biochemically; immunhistochemically expression of FGF23, VDR, RANK was investigated. In microtomographic evaluations, the amount of ossification, bone volume and mineral densities were found to be statistically significantly higher especially in the 4th and 8th weeks in the groups that calcitriol was attached. In histological and macroscopic examinations, it was found that the defect area was filled with new cells and the defect area shrank more than other groups in calcitriol+chitosan+PRP group. In immunohistochemical studies, the expression of RANK, FGF23, VDR antibodies was more and more intense in the calcitriol+chitosan+PRP group compared to other groups. Biochemically, it was observed that vitamin D level increased and wieght decreased only in calcitriol+chitosan+PRP group. As a result, it has been observed that adding vitamin D, PRP, chitosan formulation to the defect area positively contributed to the repair of bone defects.
