Abstract (EN)
Breast cancer is the second most common type of cancer among women in the USA and it is one of the most studied cancer type in the world. As an alternative for the standard treatment used in breast cancer there is a need for agents which are more potent and have less toxicity. Deguelin is an agent among the "promising" agent that has positive foreknowledge in the literature, although it has only been experimented on a few cell lines. The aim of this study wasto experiment the activity of Deguelin, which might be anti-cancer agent, on breast cancer cell lines and to compare its toxicity to the medicine used in the standard treatment. The research was planned to be made on the breast cancer cell lines representing this nature. Once the effective concentration for each agent on the cell lines was obtained, the next steps of our study were studied with this concentration.The effectivity of the two agents (Doksorubisin and Paklitaksel) used in the standard treatment was compared to Deguelin. IC50 doses were determined for Deguelin, Paklitaksel and Doksorubisin by cell vitality tests on the breast cancer cell lines.These doses were handled effectively and used as treatment doses. Flow cytometric analyzes (apoptosis,cell cycle), Oxidative Stress Index (OSI),SOD analysis was be done to treated cells and the effects of agentswas be compared by obtaining evidences related to the tumor cell line 3d growth such as Colony Formation,Migration and Angiogenesis Assay.Statistical analyzes were executed in the SigmaStat 3.5 programme by using the ANOVA and Holm-Sidak tests. In the study, it was observed that Deguelin had similar characteristics to the agents used in the treatment of breast cancer cells, and the anticancer effect could be reduced by the spread of cancer cells.
