Measurement of asymmetric dimethylarginine, nitric oxide levels, and total antioxidant capacity in experimental diabetic rats.

Abstract (EN)

In this study, it is aimed to measure the Asymetric Dimethylarginine, Nitric Oxide and Total Antioxidant Capacity levels in rats with experimental diabetes. 8-10 weeks aged, 24 male Sprague-Dawley rats were employed in the study. Rats were observed at Adnan Menderes Univercity Faculty of Veterinary Laboratory of Experiment Animals in light (12 hours day light and 12 hours night), at 22ºC and fed with adequate food and water.Animals are divided into two groups which are called control (n=12) and experimental (n=12) Before the study hunger blood sugar of the animals were measured and it was determinated 112±9.50 mg/dl. To form experimental diabetes, single dose of 50 mg/kg intraperitoneal streptozotocin in 0.01 M phosphate citrate buffer (pH 4,5) were injected to the experimental group (n=12). Hunger blood sugar rates were measured 21 days after the injection. Intra core blood is taken into tubes under ether anesthesia after experimental groups?s hunger blood sugar were measured 425±50 mg/dl. Serum and plasma was divided after tubes were santrifuged with 3000 rpm for 10 minutes and samples are kept in -20ºC until analysis.The ADMA for control and diabetic rats is found 5,59±0.75 ng/ml and 7,26±0,86 ng/ml, the NO for control and diabetic rats is found 62.28±10.74 µM/ml ve 30.42±2.48µM/ml, the MDA for control and diabetic rats is found 26.38±1.94 ?mol/L and 42.2±1.24 ?mol/L and the TAK for control and diabetic rats is found 1,01±0.06 mmol trolox equivalent/L ve 0.87± 0,03 mmol trolox equivalent/L respectively.In this study, according to the findings of the experimental diabetes model which was induced by using the increasing glucose levels both increases plasma MDA values and leads to oxidative stress, lipid peroxidation and a decrease in total antioxidant capacity. In addition, hyperglycemia by affecting ADMA metabolism causes an increase in serum ADMA levels and parallelly causes a decrease in NO levels.Key words: Diabetes, Streptozotocin, Asymetric Dimethylarginine, Nitric Oxide, Total Antioxidant Capacity.
