Effect of lactobacillus acidophilus on serotonin in rats with experimental ulcerative collitis

Abstract (EN)
Objective: Ulcerative Colitis is a structural disorder in the colonic mucosa as a result of recurrent attacks of inflammation. Today, probiotics are also used to support the drug treatment of ulcerative colitis. Probiotics are microorganisms that positively affect human health. In our study, it was aimed to investigate the effect of lactobacillus acidophilus on serotonin in experimental ulcerative colitis. Method: In our study, 40 adult male rats were used. Our experimental groups; were divided into 5 groups as control, experimental colitis, sham, probiotic treatment and probiotic preservative group. Elisa was evaluated for the analysis of serotonin protein level in the colon tissue and immunohistochemically with rabbit anti 5-HT antibody for the expression of serotonin. The obtained data were analyzed statistically. Results: In the experimental colitis, treatment and preventive group, it was determined that the release and intensity of serotonin in the colon was triggered and there was a significant increase. There is a statistically significant difference especially in the experimental colitis group. While the serotonin ng/mg protein level in the colon increased significantly in the experimental colitis group (1.98±0.52) compared to the other groups, the prophylactic (1.77±0.51) and treatment (1.72±0.26) groups had a significant increase in serotonin ng levels. /mg protein level was significantly increased compared to the control (1.38±0.42) and sham (1.48±0.50) groups (p=0.01, p<0.05). Serotonin expression in the colon increased significantly in the experimental colitis (0.15±0.02) group compared to the other groups, while it was significantly higher in the protective (0.12±0.02) and treatment (0.12±0.01) groups in the control (0.02± 0.03) and sham (0.04±0.02) groups (p=0.01, p<0.05). Conclusion: In our study, significant results were obtained for the colon in the protective and therapeutic effect of probiotics. We think that the relationship between probiotics and the amount of serotonin in the colon will contribute to the literature.
