Abstract (EN)
Aim: In this study, it was aimed to measure the apelin, dynamic thiol-disulfide balance and albuminuria levels in patients with type 2 diabetes, and to examine the role of the measured parameters in the pathogenesis of the disease. Material and Methods: For this purpose, 87 patients with type 2 diabetes mellitus and 24 healthy control groups equal in age and gender were included in the study. Morning fasting blood was taken from all individuals into serum and plasma tubes. Samples were then centrifuged at 4000 rpm for 10 minutes, and serum and plasma were portioned and stored at -80 ° C. Total and native thiol tests in plasma samples were studied with the colorimetric method developed by Erel et al. Disulfide levels were calculated. It was studied on hemogram and HbA1c whole blood samples. Biochemistry parameters (Glucose, BUN, creatinine, AST, ALT, bicarbonate, sodium, potassium, chlorine, phosphorus, alkaline phosphatase, albumin, total protein, uric acid, albumin and creatinine in spot urine) levels were measured in the autoanalysis by spectrophotometric method. C-reactive protein (CRP) levels were studied by nephelometric method. Serum parathormone, 25-(OH) vitamin D and ferritin levels were measured. GFR levels were calculated according to the creatinine based CKD-EPI formula. Apelin levels were studied by ELISA method. The data obtained were statistically analyzed and discussed. Results: As a result of the study, native and total thiol levels were significantly lower in the diabetic patient group compared to the control group (p = 0.001 for both). In contrast, calculated disulfide levels were similar between the patient and control groups (p = 0.182, p = 0.119). Serum apelin levels were significantly lower in the diabetic patient group compared to the control group (p XIII <0.001). Negative correlation was found between native thiol, total thiol and apelin levels and age. A significant and negative correlation was found between native thiol, total thiol, apelin levels and glucose, HbA1c and albuminuria. In addition, it showed a significant and positive correlation with native thiol, total thiol, apelin levels and GFR. There was no correlation between disulfide and age and GFR. Conclusion: As a result, colorimetric measurement of dynamic thiol levels can contribute to the diagnosis and follow-up of the disease as a marker due to its easy application in routine clinical biochemistry laboratories and its relationship with disease severity in patients with CKD. The Apelin / APJ system may play a role in the pathogenesis of diabetes and its complications. Therefore, further studies are needed in animals and even humans.
