Investigation of the relationship between some gene polymorphisms in diabetic obese and non-diabetic patients

Abstract (EN)

Diabetes mellitus is an endocrine and metabolism disorder characterized by carbohydrate, lipid and protein metabolism disorders, the most typical symptom of which is hyperglycemia, which is caused by the complete or partial lack of secretion of insulin hormone or insulin effect in the cells of the Langerhans chambers of the pancreas. While the most typical clinical symptoms in diabetes mellitus patients are polydipsia, polyphagia and polyuria, capillary membrane changes and accelerated arteriosclerosis formation are observed. Patients are usually obese or overweight [Body mass index (BMI)> 25 kg / m2. When the genetic relationship of the cause of diabetes and obesity is examined, many genes found to be responsible were found. The purpose of our study; To examine the relationship between gene polymorphisms in terms of case groups of FTO, FABP2 and TCF7L2, which are thought to have an effect on both diabetes and obesity. In our research; While 82 diabetic obese patients and 82 non-diabetic obese patients were used, 70 obese and non-diabetic volunteers were included in the control group. The gene polymorphism in our groups was performed by DNA isolation and Real-Time PCR method, and the results were evaluated by SPSS. As a result of our research; FTO-FABP2 genes and alleles showed statistically high significance in our group comparisons. In addition, statistically high significance was found in the comparison of these genes and alleles with BMI, age, glucose and HbA1C parameters. These genes have been found to be associated with both diabetes and obesity. Statistically sufficient clarity has not been achieved for TCF7L2 gene, but it has been shown to have to be connection on diabetes. In order to reach detailed and clear results in terms of the related gene, it is recommended to conduct other studies with more samples.
