The genetic diversity analysis of different bean (Phaseolus vulgaris) cultivated genotypes in Turkey using DNA-based moleculer methods

Abstract (EN)
It is known that there are around 230 species of beans in the world and 20 of them are used extensively in human nutrition and P. vulgaris is the most produced species. In this study, the DNA-based molecular markers of 38 beans genotypes obtained from Eskişehir Agricultural Research Institute an important database has been created for rapid recovery of new varıetıes which are both resistant to determined diseases and with high efficiency in the future improvement studies with ISSR (Inter Simple Sequence Repeat Polymorphism), RAPD (Random Amplified Polymorphic DNA) and ITS (Internal Transcribed Spacer) markers. Ph.Corh cultivars belonging to different bean species were used as external groups in the analyzes. The band analysis of the PCR products of the selected markers was performed by Phoretix1DPro program. By using the data obtained by the binary method, Jaccard similarity matrix was obtained by UPGMA method. With the Newick format obtained from matrix data, phylogenetic tree was created for each method in Mega 6.0 program. As a result of distance matrix data obtained by tree obtained by ISSR primer, UI1140 and Novanbey (distance matrix: 0.946) genotypes were found to be the most distant individuals and the drawn tree with RAPD primers, Cardinal Yozgat Barbunyası and 17SBVD20 (distance matrix: 0.805) genotypes were identified as the most distant individuals. The phylogenetic tree obtained by the ITS method was not evaluated because the distinction of the out-group was not clear. Data obtained from this study performed using genotypes that genetic relationship wasn't analyzed have got important data qualification to develop new more resistant and fruitful varieties in future breeding studies.
