Abstract (EN)
SUMMARYAlcohol addicition is one of the leading reasons of liver diseases in many countries.Hepatic steatotis, alcoholic hepatitis, hepatic fibrosis and cirrhosis can be mentionedamong the diseases resulting due to alcohol addiction. In recent years, studies havebeen focused on effect of oxidant stres, in the process of pathological changes andmentioned diseases caused by alcohol. In many steps of alcohol metabolismdependent of production of reactive oxgen species (ROS) that is occured alcoholinduced pro-oksidan stres. Lipid peroxidation and membran damage is developed byROS. In our body, against the harmful effects of ROS there is an antioksidanmechanism. Antioksidants which are break the lipid peroxidation chains reactionsconstitue an important part of the antioksidant defense. Vitamine E is one of theimportant antioksidant. Matrix metalloproteinases (MMPs) are an enzym group ofthat degrade extracelluler matrixs (ECM). MMPs also called matrixins. MMPs areZn+2 and Ca+2dependent endopeptidases that breakdown the ECM. The aim ofpresent study, we evaluate wheter the vitamine E treatment could have a protectiveeffect against oxidative stress and pro-MMP-9 and the oxidative stress and role ofpro-MMP-9 in alcohol induced liver injury in rats or not.In this study, twenty five male Sprauge-Dawley rats were divided fourgroups: 1) Control group: recieved saline, intragastrically (2 ml/day) througout thirtydays. 2) Ethanol (EtOH) group: received EtOH (1 ml/day, 80% v/v), intragastricallyand saline througout thirty days. 3) Vitamine E (VitE) group: recieved VitE (100mg/kg/day) with saline that total volume is 1 ml througout thirty days. 4)VitE+EtOH: VitE was introduced 2 h before EtOH administration (1 ml 80% v/v)througout thirty days.Malondialdehit (MDA) levels as an indicator of lipid peroxidation, content ofprotein corbonyl as an indicator of protein oxidation, the levels of glutatyon (GSH)and proetin sulphydril groups (SH) and superoksid dismutase (SOD) activities asindicators of antioksidant capacity and proMMP-9 levels have been estimated in livertissue.PDF created with pdfFactory trial version www.pdffactory.comAs a result of these measurement, it has been shown that increased tissueMDA, protein carbonyl groups, proMMP-9 and decreased GSH, SH levels anddecreased SOD activities in EtOH group were found when compared to controlgroup. Decreased tissue GSH, SH levels and increased SOD activities in EtOH+VitEgroup were also found when compared to EtOH group. The SOD activities were notsignificantly different from other groups.In conclusion, alcohol effect betwen oksidant and antioksidant balance inliver and it is increased lipid peroxidation. Alcohol induced oxidative stressincreased significantly proMMP-9 levels of liver tissue and VitE decreased oxidativestress and we think that decreased proMMP-9 levels are dependent decreasing ofoxidative stress.Key Words: Oxidative stress, Ethanol, proMMP-9, Vitamine EPDF created with pdfFactory trial version www.pdffactory.com
