Evaluation of serum anti-mullerian hormone concentration for determination of ovarium reserve in cats

Abstract (EN)

In this thesis study, it was aimed to determine the correlation between ovarian follicle numbers and serum anti-Müllerian hormone in 1-2 year old cats in the estrus and interestrus stages of the sexual cycle. Vaginal cytology was performed and group Ӏ (n=15) estrus and group ӀӀ (n=12) interestrus group were formed. The cats included in the study were first subjected to physical examination, hemogram, and ultrasound examination. Unhealthy cats were not included in the study. Follicles removed by the operation were first kept in 10% formaldehyde. Then, the hematoxylin-eosin staining method was applied to the first of the 5-6 µm serial sections to be taken from the prepared blocks, and it was determined whether there was any lesion in the microscopic examination. The average number of follicles was determined by counting the follicles in 5 sections in each ovary. Follicles are classified as primordial, primary, secondary, early antral and antral follicles. Serum AMH, progesterone and estrogen concentrations were measured by ELISA method. As a result, the average number of follicles was calculated by counting the follicles in both ovaries of the groups included in the study. The number of primordial follicles was 213,8 ± 99,9 and 168,3 ± 136,5 in the estrus and interestrus groups, respectively. The number of primary follicles was determined as 75,0 ± 46,2 and 57,7 ± 29,0 in the estrus and interestrus groups, respectively. The number of secondary follicles was observed as 52,3 ± 50,9 and 40,9 ± 25,2 in estrus and interestrus groups, respectively. The number of early antral follicles was 1,5 ± 2,5 and 1,3 ± 1,3 in the estrus and interestrus groups, respectively. The number of antral follicles was found to be 2,4 ± 1,8 and 3,1 ± 1,8 in the estrus and interestrus groups, respectively. The number of preantral follicles was found to be 341,1 ± 172,9 and 266,9 ± 158,4 in the estrus and interestrus groups, respectively. There was no statistical difference between estrus and interestrus groups in terms of follicle numbers. The number of corpus luteum in the interestrus and estrus groups was found to be 3,0 ± 1,4 and 3,2 ± 2,3, respectively. Serum AMH levels were recorded as 1,4 ± 0,5 ng/ml and 3,1 ± 1,2 ng/ml in the estrus and interestrus groups, respectively. Serum progesterone concentration was 0,5 ± 0,1 ng/ml and 0,87 ± 0,1 ng/ml in the estrus and interestrus groups, respectively. Serum estrogen levels were 58,4 ± 13,6 pg/ml and 42,3 ± 11,4 pg/ml in the estrus and interestrus groups, respectively. It was determined that there was a significant positive correlation between serum AMH concentration of cats in the estrus group and primordial and preantral follicle numbers. As a result, although the follicle numbers of the groups were the same, serum AMH levels differed.
