Abstract (EN)

In this work, the effect of combination of different medium in some cucumbers (Cucumis sativus L.) genotype by Gynogenesis (ovary culture) on embryo formation and haploid plants were investigated. These studies worked on Kuzey Agripark Bitki, Araştırma ve Biyolojik Mücadele San. ve Tic. Ltd. Şti. station on 2014-2015 years. In this survey, three combination of different culture medium was modified fixed rate by plant growth regulator for embryo induction and plant regeneration. Effect of mediums, genotype and plant age were examined weekly. According to the findings seen clearly the importance of the medium, that conclusion has reached MS + 1:10; 2,4- D: Kinetin (MSB ) is the most effective incentives embryo, while the most effective regeneration medium, MS + 1:4; NAA:BAP (MSR). All genotypes, have created embryos on MSB medium and plant regeneration was carried out in MSR medium. Total 273 embryos obtained, 137 plant regeneration were occured. Total applied by the number of ovary; % 84.26 embryo formation rate and % 42.28 plant regeneration rate was obtained. Genotype effect on research has shown clearly and the most successful genotypes No. 583 genotypes. has been identified that the embryo and the resulting plant. Second genotype was No 584 genotypes, while No 550 genotypes showed limited result has emerged very limited results. But, plant was obtained from No 550 genotypes. Depending on the increasing age of the plant , the ratio of the resulting embryo and the plant has experienced a serious decline . The donor plants grown approximately 2 months of age period the embryos initially % 213.58 ratio is decreased to % 19.75 in the last experiment established. The plant regeneration rate decreased from % 132.10 to % 4.94 level and it is showed that importance of genotype age. The study, 114 of the 137plants ploidy level was determined , which 87 of them haploid , 9 of them diploid and 18 of them miksoploid was observed.
