Cytotoxicity effects of ellagic acid on different human breast cancer cells

Abstract (EN)

In this study, the SKBR-3 and MDA-MB-231 which are human breast cancer cell lines were investigated on the cytotoxic effect of ellagic acid. For this purpose, 10 different dose ellagic acid (5?M, 10?M, 20?M, 30?M, 40?M, 50?M, 100?M, 150 um, 200?M, 250?M) is applied to cells for 72 and 96 hours.According to the results of the MTT cytotoxicity tests, the number of living cells is decreased, the number of dead cells is increased by depend on time and dose.After microscopy analysis via transmission electron microscope (TEM) and fluorescence microscope performed, half maximal effective concentration (EC50) was determined for the cells being tested with MTT cytotoxicity assay. Upon ellagic acid addition to cells at 72h and 96h after, the experiment resulted with loss in cell volume, nucleus condensation and apoptotic body formation.Effect of ellagic acid on SKBR-3 and MDA-MB-231 cells was investigated compared with respect to dosage concentration of ellagic acid and time. As a result, ellagic acid had cytotoxic effects on cells by time and dose dependend manner.Keywords: Cancer, Ellagic acid, MTT, TEM, Fluorescence Microscopy
