Determination of prokaryotic diversity in different salt deposit samples

Abstract (EN)

In this study, salt deposit samples obtained from salt mines in Çankırı, Nevşehir, Kırşehir, Yozgat, Erzurum, Kars provinces, were analyzed for determination cultivable prokaryotic diversity. 16 halophilic prokaryotic isolates have been collected from these salt deposits, by using different media. DNA have been extracted from 11 of the isolates and by using the Polymerase Chain Reaction (PCR), Archaea and Bacteria domain were differantiated. 3 of the isolates were determined as Archaea while the other 8 isolate were determined as Bacteria domain. The 16S rDNA analyses has shown that Archaea members were in Halobacterium genus, while the Bacteria members were belong to Oceanobacillus sp. and Oceanobacillus iheyensis. Microbial community of the samples have been investigated by fluorescent in situ hybridization (FISH) and DAPI staining techniques.
