Abstract (EN)

In this study, Ferula orientalis L. and endemic species for Turkey Ferula mervynii M. Sağıroğlu & H. Duman plants collected from Erzurum, in vitro antidiabetic, anticholinesterase and cytotoxic activities were tested on dichloromethane and methanol extracts obtained from the roots of both species and the root dichloromethane extracts were found to be active. Isolation studies were performed on active extracts using chromatographic methods and obtained molecule's structures were elucidated. The test systems were applied to the compounds with sufficient amount. All substances were isolated for the first time from Ferula mervynii. Six new sesquiterpene esters; 6-acetyl-8-t-cinnamoyl-antakyatriol (FM-2), 6-acetyl-8-t-cinnamoyl-shiromodiol (FM-4), 8-t-cinnamoyl-tovarol (FM-7), 6-acetoxy-8-t-cinnamoyloxy-nor-β-1(S),2-epoxy-elemane-4-on (FM-8), 6β-acetoxy-8α-t-cinnamoyloxy-nor-epi-elemane-4-on (FM-19), 6-acetyl-8-t-cinnamoyl-3-epi-antakyatriol (FM-20), three known sesquiterpene esters; 6-acetyl-8-t-cinnamoyl-antakyatriol (FM-1), 6-acetyl-8-benzoyl-tovarol (FM-5), ferutinin (FM-11), two known phenylpropanoids; crocaton (FM-3), 1-hydroxy-1-(3'-methoxy-4'-5'-methylenedioxy)phenylpropane (FM-12) and one known acetylenic compound; falcarindiol (FM-13), totally 12 molecules were isolated. Eleven known compounds were isolated from the root dichloromethane extract of Ferula orientalis and identified using spectroscopic methods. Six molecules; ferutinin (R-2), 8-p-hydroxy-benzoyl tovarol (R-3), epoxyjaeschkeanadiol vanillate (R-5), epoxyjaeschkeanadiol p-hydroxy benzoate (R-6), 6-β-p-hydroxybenzoyloxy-germacra-1(10),4-diene (R-7) and 8-vanilloyl tovarol (R-11) were obtained like another isolation study on F. orientalis. In addition, leucoferin (R-9), teferin (R-14), teferidine (R-15), 8-O-angeloyl tovarol (R-16) and chimgin (R-25) were determined for the first time from this species. In this study; on α-amylase and α-glucosidase enzymes for antidiabetic activity, acetylcholinesterase and buthylcholinesterase enzymes for anticholinesterase activity, colon (COLO 205), leukemia (K-562), breast (MCF-7) and human umbilical vein endothelial (HUVEC) cell lines for cytotoxic activity were used for biological activity test systems.
