Therapeutic effect of carbon monoxide on Glioblastoma multiforme compared with temozolomid-cell culture study

Abstract (EN)

Although carbon monoxide (CO) has been accused of being an inhaler toxic gas, recently it was acknowledged as an intracellular mediator in different pathways like "heme" metabolism or apoptosis. CO has been shown as an anti-inflammatory, anti-hypertensive, anti-oksidant and anti-tumoral mediator via endogenously produced by heme oxygenase (HO) enzyme in the body. In the light of this information, carbon monoxide releasing molecules (CORMs) were produced for controlled release of CO and making use of beneficial effects of CO exogenously. CORMs were especially used in cancer researches like breast, prostate, colon, cervix, pancreas and skin cancers in the literature and positive effects of CO has been shown in the treatment of these cancers. Although there are a lot of studies conducted on different cancers; there are no studies on central nervous system (CNS) tumors, yet. In this dissertation study, we aimed to study effects of CO released via CORMs on glioblastoma multiforme (GBM) cells which is one of the deadliest tumors. Effects of CORMs on GBM cells were investigated in medical biology laboratory by ESOGÜ Neurosurgery and Medical Biology Departments. In conclusion, CORM-2 50 µM decreased cell proliferation more than temozolomide (TMZ) at 24th hour which is accepted as the most important hour for the proliferation. However, different doses of neither CORM-2 nor CORM-3 had influence on the viability as TMZ. As a result, CORM-2 50 µM might be a successful alternative drug for GBM in future in vivo studies.
