Investigation of the effect of viniferin on oxidative stress in hela cell line

Abstract (EN)

Throughout the known history, the most common problem that threatens the life of a living person and is common has been cancer. Cervical cancer is the fourth most common cancer in women worldwide after breast cancer (2.1 million cases), colorectal cancer (0.8 million) and lung cancer (0.7 million). The antioxidant activity of ε-Viniferin aims to prevent both chemical-induced cancer and oxidative damage by preventing the onset and progression of cancer. In this thesis, it is aimed to investigate the effects of ε-Viniferin on oxidative stress in HeLa cell series. In order to determine the cytotoxic effects of cisplatin (CDDP) and ε-Viniferin on the HeLa cell line, the MTT method was used, as a result of which the IC50 value for CDDP was calculated as 28 µM, and the IC50 value for ε-Viniferin was calculated as 21 µM. Significant difference in Total Antioxidant Levels (TAS) and Total Oxidant Levels (TOS) in the combined groups calculated with IC50 values, among the synergistic groups selected according to the combination index 20% for TAS (5.6 µM CDDP + 4.2 µM VIN), 10% for TOS (2.8). µM CDDP + 2.1 µM VIN) groups, whereas Oxidative Stress Indexes (OSI) were calculated greater than 1 at all doses applied. Superoxide Dismutase (SOD) values of the 20% and 10% combined groups, which we think have a synergistic effect, are lower than the control (1,550 ± 0.10 U/mL), and Reduced Glutathione (GSH) values compared to the control (1.378 ± 0.09 μM) lower than the control and Malondialdehyde (MDA) values were higher than the control (0.461 ± 0.000 μM). As a result, according to the combination index, when the synergistic combination groups are compared with CDDP given alone, a minor level of oxidative stress inhibitory property can be observed, but according to the statistics made compared to the control, although ε-Viniferin partially reduces the oxidative stress caused by CDDP, we expect it. It has been observed that it has no effect on reducing cell viability at the level of as expected.
