Özet (EN)

Objective: Hepatocyte Growth Factor (HGF) is known to increase expression as in other cancer types. This study aimed to investigate expression at the transcriptional level of HGF expression. Because of this, we examined the presence of mutation in the hepatocellular carcinoma cells in the region of 30 reprocessed deoxyadenosine in the HGF promoter and found that it was related to high HGF levels Method: Two hepatocellular carcinoma cell lines and two human genomic DNA lines were used in this study. Genomic DNAs were obtained with the aid of kit and region specific primers were designed and amplified. Next, bi-directional Sequencing was done. Chromatogram data showed deoxy adenosine levels in the region. Subsequently, the present HGF expressions in these cells were compared with our data Results: When HGF mutations were normal in some hepatocellular carcinoma cells, shortening in DATE region was observed in nascent cell lines, and it was determined that the number of deoxyadenosine in the DATE region was normal in cells with high autocrine HGF expression Conclusion: HGF is responsible for the activation of autocrine HGF in the DATE region due to its transcription in some cancer types. Our results suggest that methylation levels should be examined in cell lines where the DATE region is normal and shortened in order to understand the role of HGF DATE region in expression
