Abstract (EN)

Carbonic anhydrases (CAs), catalyze a reversible reaction of carbon dioxide hydration and dehydration, thus playing a vital role in metabolic processes by providing acid-base and ion regulation. CA VII is expressed in the colon, liver, skeletal muscle and brain. CA VII expression in tumors is much lower compared to other isoforms, with the highest expression observed in colorectal adenocarcinoma, thyroid carcinoma. In colorectal adenocarcinoma, CA VII upregulation is thought to serve as a marker for poor prognosis. Cancer cells, due to their rapidly proliferating nature, rapidly consume the nutrients and oxygen in the environment, making their microenviroment more hypoxic. Hypoxic microenvironment is an important pathway that can cause changes in the regulation of many genes in cancer. In this study, the expression of CA VII gene in different cancer cell lines and its changes in hypoxic conditions were investigated at the mRNA and protein levels. Our studies continued in the Chemical hypoxia model with CoCl2 and sodium sulfide. The expression of CA VII gene in colon cancer cell lines (SW480 and HT-29), healthy human endothelial cells (HUVEC), Liver cell line (Hep3B), prostate cancer cell lines (PC3 and LnCaP) was primarily examined at the mRNA level. CA VII mRNA level was detected in all cell lines except HUVEC cell line. When all the results were evaluated within the scope of our study, it was determined that both mRNA and protein levels decreased statistically significantly in the HT-29 cell line. It was determined that CA VII level increased in both mRNA and protein levels in LnCap cells, which is one of the selected prostate cancer models. In a second arm of our study, experimental studies were also planned in C.elegans, which is a good model organism in molecular studies. In this context, the expression of CA VII-like genes (CAH 3, 4 and 5) and the change in mRNA level under hypoxic conditions in C.elagans were studied. mRNA expression of selected genes is shown in C.elegans.
