Distribution of the HIV-1 resistance-conferring Alleles CCR5-Δ32, CCR2-64I ve SDF1-3'A in turkish population

Abstract (EN)

AIDS caused by Human Immunodeficiency Virus-1 (HIV-1) has continued to spread rapidly throughout the world. Despite all conducted studies, no treatment that can eradicate the HIV virus completely has been found. All of the vaccines developed against the virus failed since the virus is capable of changing its genetic structure. Besides, it has been shown that CCR5-Δ32, CCR2-64I, and SDF1-3'A genetic polymorphisms occurring on CCR5, CCR2, and SDF1 genes in human respectively provide effects that confer resistance to HIV infection and decelerate the progression of AIDS. In this study, genomic DNAs obtained from blood samples with salting-out method from 205 healthy volunteers of Sinop province were investigated by PCR and PCR-RFLP methods. The genetic liability of Turkish population was evaluated by calculating the allele frequencies of CCR5-Δ32, CCR2-64 and SDF1-3'A polymorphisms. Allele frequencies of these genes in the population were found as 3.17%, 17.80% and 27.07% respectively for CCR5-Δ32, CCR2-64I and SDF1-3'A and all the observed genotype frequencies were in accordance with Hardy-Weinberg equilibrium. CCR5-Δ32 allele frequency is relatively lower compared to European countries but higher compared to Asian and African countries. CCR2-64I and SDF1-3'A allele frequencies coincide with many countries and ethnicities in different continents. Allele frequencies of three polymorphisms investigated are similar with neighbour countries Greece, Crete, Greek Cypriots, and Bulgaria. The medical world has begun to focus on the diagnosis and treatment applications based on the individual genetic structure. Investigation of genetic polymorphisms is important in terms of identifying high-risk individuals for HIV-1 infection in Turkish population and potential of future individualization of HAART therapy.
