Abstract (EN)

In the presented thesis study, a new analytical method based on the use of a boron-doped diamond electrode for the electrochemical determination of Brentuximab vedotin, an anticancer drug used in the treatment of Hodgkin's lymphoma, was developed. In this developed electroanalytical technique, it was observed that Brentuximab vedotin produced an irreversible wave at approximately +1.35 V in a basic medium on a boron-doped diamond electrode that had been electrochemically activated in a sulfuric acid environment using the cyclic voltammetry technique. In the technique developed for the quantitative determination of Brentuximab vedotin, the linear equation in the concentration range of 9.99 μg/mL to 49.95 μg/mL on a boron-doped diamond electrode in a Britton-Robinson (pH =8.0) supporting electrolyte medium was determined as Ip(μA)=0.026 C[BV] (μg/mL) -0.269, r=0.997 (n=9). The lowest detection limit of the developed method was calculated as 0.98 μg/mL. In addition, the accuracy study of the developed method was carried out by applying a standard addition-recovery process to urine samples. As a result of the analysis, a high recovery result of 94.25% was obtained for Brentuximab vedotin found in urine samples. This has shown that the electrochemical method we developed for the determination of Brentuximab vedotin in urine can produce results with high accuracy.
