The effects of different media on node and shoot tip cultures of Hyssopus officinalis L. plant at in vitro conditions

This research was carried out to examine the effects of different combinations of plant growth regulators on in vitro micropropagation of Hysoppus officinalis L., being an aromatic and medicinal herb. Node and shoot tip explants were obtained from in vitro germinated sterile seeds. Explants were cultured on MS medium containing different concentrations and combinations of various plant growth regulators (1,0- 2,0- 3,0 mg/l BAP and 0,1- 0,5- 1 mg/l NAA and IBA). The longest shoot from shoot tip explants was observed in the MS medium supplemented with 1,0 mg/l BAP (1.84 cm). The highest amount of leaf formation from shoot tip explants was obtained in the MS medium supplemented with 1,0 mg/l BAP, 1,0 mg/l BAP - 1,0 mg/l NAA getting the numbers of 16.00– 15.76, respectively. The highest amount of leaf formation from node explants was obtained in the MS medium supplemented with 3,0 mg/l BAP (18.63 numbers). The shoots were transfered to 9 different root media. The highest rootting percentage was observed in the MS medium supplemented with 1 mg/l NAA (% 78,3) and root development were not observed in 1/2 MS medium without hormone supplement. The obtained rooted plantlets were successfully acclimatized in soil.
