The investigation of cytotoxic effects of dinuclear copper-hydrazone complex

Abstract (EN)

In this study, the cytotoxic effects of the binuclear Copper - Hydrazone Complex on human lung cancer cell lines (A549 and H460) and healthy human cells on human lung bronchial epithelial cells (BEAS-2B) were investigated by using (MTT) assay depending on concentration (0.01; 0.1; 1; 10 and 20 µM) and time depend manner. In addition, the antiviral effect of the binuclear Copper - Hydrazone Complex on Poliovirus Type 1 grown in Vero cell culture, antibacterial effect on Bacillus subtilis, Enterococcus hirae, Staphylococcus aureus, Escherichia coli and Pseudomonas aeruginosa bacteria was determined using Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBK) tests. The IC50 values of the Copper - Hydrazone Complex in cell A549 at 24 and 48 hours were 4 and 2 µM, respectively; for the H460 cell, they were calculated as 8 and 5 µM, respectively. IC50 values in BEAS-2B cells were calculated as 11 and 7 µM for 24 and 48 hours. It was determined that 0.01; 0.1; 1; 10 and 20 µM carboplatin doses used as positive control in the study had very low toxicity on both cell lines in both 24 hour and 48 hour applications, and the IC50 value could not be calculated. As a result, it was determined that the Copper - Hydrazone Complex, which was tested for the first time in human lung cancer cell lines, suppressed cell proliferation in both A549 and H460 cells, depending on the dose increase, especially from the 1 µM dose. It was observed that this effect increased depending on the dose, but did not change depending on the time. In addition, any antiviral activity of the binuclear Copper - Hydrazone Complex on Poliovirus Type 1 at the concentrations mentioned could not be determined. As a result of antibacterial activity experiments, it was determined that the compound did not have antibacterial activity on Escherichia coli and Pseudomonas aeruginosa. MIC and MBK values, respectively, were 31.25-62.5 for Bacillus subtilis; 62.5- 250 µM for Enterococcus hirae and 62.5- 500 µM for Staphylococcus aureus.
