Determination of some probiotic properties of bacteria isolated from cow colostrum milk

Abstract (EN)
In this study, the probiotic properties of 11 Lactobacillus spp isolates isolated from cow colostrum were investigated. As a result of the studies; it was determined that Lactobacillus spp. OO1, OO3, OO4 and TO4 isolates showed high tolerance against 0.4 %phenol. Isolated Lactobacillus spp. isolates showed sensitivity to low pH condition (pH 2.0), while they maintained their viability at pH 3, pH 4 and pH 7. Lactobacillus spp. isolate's average viability in 0.3% and 0.5% bile environments were determined as 8.56 and 8.04 log cfu/mL respectively. The antimicrobial effect of metabolites obtained from Lactobacillus spp isolates on food and intestinal pathogens was investigated and results revealed that isolates have antimicrobial activity against test bacteria Escherichia coli ATCC 35218 (3.04-17.48 mm), E. coli ATCC 11229 (2.64-17.16 mm) Salmonella enteritidis ATCC 13076 (2.24-11.56 mm), Bacillus cereus RSKK 863 and Staphylococcus aureus ATCC 25923 (6.98-20.18 mm). Lactobacillus spp. isolates were tested using Lactobacillus isolates were tested for their susceptibility to ampicillin, ofloxacin, erythromycin, chloramphenicol, netilmicin, gentamicin, and polymyxin antibiotics. While the OO4 strain showed resistance to ampicillin and gentamicin, the TO4 strain exhibited resistance to gentamicin. The production of EPS believed to be effective in the aggregation and hydrophobic activities of probiotic strains was determined between 205.91 and 372.36 mg/L. While autoaggregation activity was observed in Lactobacillus spp. isolates at a rate of 9-31%, low levels of coaggregation were observed with test bacteria S. aureus ATCC 25923 and E. coli ATCC 35218. Lactobacillus spp. isolates exhibited hydrophobicity values ranging from 13.5% to 94.80%, 1.15% to 85%, and 1.6% to 84.8%, respectively. Strains TO3, TO4, and OO4, with high EPS production, showed probiotic properties such as aggregation, hydrophobicity, antimicrobial activity, and tolerance to gastrointestinal conditions. The hemolytic activity of these strains was determined as γ-hemolytic. The TO4 isolate, which showed the highest activity among the isolates whose probiotic properties were investigated, showed 99.86% similarity to the Limosilactobacillus fermentum species as a result of 16S rDNA sequence analysis.
