Abstract (EN)

Exosomes, one of the extracellular vesicles, provide molecular information and intercellular communication between cells by containing many essential molecules such as various proteins, lipids, mRNA, metabolic enzymes and miRNA. Exosomes have emerged as important mediators of intercellular communication in cancer and can program the growth and spread of cancer by changing the local and systemic environment. Considering the limited number of studies in the literature on the multifaceted effects of exosomes on cancer progression and the effects of exosomes on cancer cells, in our study, exosomes were isolated and characterized by TEM, Western blot and immunofluorescence analysis. Then, proliferation/cytotoxicity, colony formation and cell migration studies were performed in SW-480 cells. In addition to these analyses, transcriptomic analyses were performed to determine the effect of exosomes on SW-480 colon cancer cells. All RNA transcripts were investigated with the bioinformatic analyses performed. Statistically significant genes were determined in the obtained results and confirmed by RT-PCR studies. Among these genes, up-regulated ATOH 8 was successfully cloned into the lentiviral-based pLKO.1-TRC vector for sh RNA studies.
