In this study, we wanted to investigate to compare the percentage of apopitosis in granulosa cells from follicular fluid use immunohistochemistry technique and ultrastructure of granulosa cells and also assessment for ROS respectively cycles in assisted reproductive technology.Our study was a prospective, nonrandomized clinical study. Dokuz Eylül University Institute of Health Sciences Ethic committee approved our application with 13 numbered decision at 19th March 2008 and also Dokuz Eylül University Ethics Committee of Clinic And Laboratory Researches, Faculty of Medicine, approved our application with 120 numbered and 28 March 2008. This study is a workshop comprising 31 IVF cycles which has applied to 11 PCOS infertile patients, 10 unexplaned infertile patients and 10 male factor infertil patients between April 2008 and December 2008, in Dokuz Eylul Unıversıty Faculty Of Medıcıne Department Of Obstetrıcs and Gynecology ,IVF Center.Follicular fluid from each follicle were separated to three groups after each case. TUNEL assay was used to detect of DNA fragmentation in granulosa cells, MDA assessment for ROS, GPx assessment for antioxidant enzyme respectively and apoptosis in granulosa cells was evaluated ultrastructurally by transmission electron microscopy.There was statistically significant difference the percentage of apopitotic cells in granulosa cells between PCOS and unexplaned infertility group. There was no statistically significant difference between the levels of MDA and GPx in follicular fluid in theree groups.With this work we have done, it was investigated immunohistochemistry and also ultrastructural apopitosis of granulosa cells which are promoting oocytes , besides the follicle fluid was investigated for the effects of oxidative stress in this situation. Consequently, in this study we broke fresh ground in PCOS and unexplaned patients for used theese 3 techniques.Keyword: Granulosa Cells, PCOS, Unexplaned Infertility, TUNEL, Apopitosis, ROS, Ultrastructure
