Abstract (EN)
Determining genotoxic effects by using in vitro cell systems in riskassessment procedures can be considerably useful. For this purpose, RAPDtechnique can be used as an initial work for assessing the genetic effects ofvarious materials. In this thesis, the effects of benzo [a] pyren, ethyl methanesulfonate and miyomysin ?C on human peripheral lymphocytes were examined byusing RAPD technique. Lymphocytes were exposured to each material for 24, 48and 72 hours and control groups were formed for comparisonas well. RAPDprofiles of exposed and non-exposed cells were compared. Differences like banddensity, increases and decreases in bands, and number of bands were observed inRAPD profiles. Depending on these RAPD profiles, it is detected that benzo [a]pyren, ethyl methane sulfonate and miyomysin ?C caused genetic damages asDNA breaks.Keywords: Genotoxicity, Human lymphocytes, DNA isolation, PCR, RAPD
