Investigation of fumagillin?s anticancerogenic effects on human breast cancer MCF7 cell lines

Abstract (EN)

Breast cancer is the most common cancer among women. Breast cancer can be metastasis without showing any symptoms. It?s shown that especially no steroidal anti-estrogen drugs can affect solely 1/3 of patients. These findings lead us alternative ways to prevent and cure to breast cancer. Recently, fumagillin, one of the secondary metabolites isolated from Aspergillus fumigatus, catches all the attention on it. Angiogenesis inhibitor, fumagillin, has strong inhibitory activities against in vivo tumor growth and metastasis in a wide variety of tumors.In this study, we investigated cytotoxic, apoptotic and antiangiogenic effects of fumagillin (25, 50, 75 and 100 µM) on MCF7 human breast adenocarcinoma cells. Cytotoxic activities of fumagillin on MCF7 cells were measured by Cell Proliferation Reagent WST-1 (stable tetrazolium salt) for 24, 48 and 72h. After 24h incubation, apoptotic and antiangiogenic effects were determined by using the Poly (ADP-Ribose) Polymerase (PARP) cleavage ELISA kit and the levels of vascular endothelial growth factor (VEGF) Human ELISA Kit, respectively. We found that fumagillin had significant time- and concentration-dependent cytotoxic effects. Especially, while fumagillin concentrations were increased, PARP level was increased, VEGF level was decreased. In conclusion, a significant cytotoxic, apoptotic, and antiangiogenic effects were observed on the MCF7 human breast cancer cell lines. Therefore, the molecular mechanisms of fumagillin should be studied as a single or combination chemotherapeutic agents, we believe these studies will provide an important contribution to the development of new chemotherapeutic and chemopreventive agents for the treatment of breast cancer.
