The effect of intravesical tarantula cubensis extract on inflamation in interstitial cystitis rat model

Introduction and Objective: It was aimed to reveal the histopathological and immunological results of intravesical treatment with Tarantula Cubensis Extract in a rat model of interstitial cystitis, thus evaluating the effect of this agent on bladder inflammation and its effectiveness as a new treatment option. Materials and Methods: A total of 30 wistar albino female rats; group 1 (control), group 2 (disease) and group 3 (treatment) were divided into 3 groups (n = 10 for each group). By applying cyclophosphamide twice a week for 2 weeks, the previously described interstitial cystitis rat model was created. In group 3, Tarantula Cubensis Extract was administered intravesically after the model was created. Histopathologically; urothelial degeneration, necrosis, ulcer, bleeding, edema, inflammation and mast cell count were evaluated. IL-6, TNF-alpha, MPO, hydroxyproline parameters were evaluated immunologically. Statistical analysis was performed with SPSS 21.0.0 program using One Way ANOVA test, Chi-square, Kruskal Wallis tests. Results: It was observed that all immunological and histological parameters were significantly lower in rats in group 1 than in other groups, and IL-6 and MPO values were significantly higher in group 2 (p<0,001). Although the mean of TNF-alpha was the highest in group 2, there was no statistically significant difference with group 3 (p=0,078). In group 1, significantly lower number of mast cells were detected from other groups (p=0,002). There was no difference between group 3 and 2 (p>0,05). When examined in terms of ulcers, no statistically significant difference was found between the three groups (p=0,087). Histopathologically, urothelial necrosis / spillage, urothelial degeneration, edema, inflammation, bleeding and fibroblast proliferation, and immunologically hydroxyproline values were found to be statistically significantly higher in group 3 (p<0,001). Conclusion: In the interstitial cystitis rat model, after intravesical instillation of Tarantula cubensis extract, IL-6, TNF-alpha and MPO levels decreased and hydroxproline level and fibroblast proliferation increased in the bladder tissue. Thus, its anti-inflammatory and fibrosis-forming effect has been demonstrated. Further studies are needed to determine the effective dose with fewer side effects.
