Determination of the antioxidant activity of nettle seed oil and investigation of its apoptotic and cytotoxic effects on lung (A549) and cervical (HeLa) cancer cells

Abstract (EN)

Cancer, which is an abnormality and occurs as a result of uncontrolled growth and metastasis, is one of the leading causes of death in the world. Lung cancer stands out as the most common type of cancer in men in the world, while the results are the third most common cancer. Cervical cancer is the third most common cancer in wowen in the world after breast cancer and colorectal cancer. In cancer treatment or support of treatment processes, medicinal plants potentially offer an option. In this thesis, the antioxidant properties of nettle (Urtica dioica) seed oil and its cytotoxic and apoptotic effects on A549 (human lung adenocarcinoma) and HeLa (cervix) systems were investigated in vitro. Antioxidant capacity was evaluated by DPPH and ABTS (TEAC) methods and 83.17% inhibition was achieved at the highest concentration in the DPPH test, but this effect was at low rates compared to the positive control. According to MTT analysis, IC50 values were determined as 0.503 mL/L and 0.5485 mL/L in A549 appearance and for 24 hours in HeLa cells, they were determined as 0,4209 ml/L, and no result was found fort he 48 hours. Annexin V test result determined the apoptosis rate as 10.05% and 13.90% in A549 cells at 24 and 48 times; 5.64% and 2.51% in HeLa cells, respectively. In caspase-3 analysis, a significant change was observed in A549, and 48.25% caspase activation was detected in HeLa appearance at 24 dates. When mitochondrial membrane potential results were examined in A549, 55.13% depolarization was detected at 48 hours. TUNEL staining for morphological imaging showed that apoptosis increased especially in A549. As a result, it was determined that Urtica dioica seed oil resulted in more necrotic cell death when A549 was grown, while apoptosis increased over time.
