In general, algae continue their lives in aquatic or semi-aquatic environments and produce food by using sunlight, thanks to some pigments in their structures. Brown and red algae, which are under the algae group, contain important compounds such as agar, carrageenan and alginic acid. Although our country ranks at the top among the Mediterranean countries in terms of the length of its sea coasts, studies on algae are few, compared to the Western Mediterranean countries. In the thesis study, first of all, ten fractions of sodium alginate from Cystoseira barbata type brown sea algae were obtained from 2 different procedures. Subsequently, in order to examine its use as an anti-adhesion barrier, a biomaterial was produced in thin film design, and the film structure with the most suitable properties was determined with its physical and chemical characterization. The usability of the films produced by selecting the fraction with the most suitable properties as an adhesion barrier was tested in in vitro culture with rat bone marrow mesenchymal stem cells (MSC) for 21 g. Mitochondrial dehydrogenase enzyme activities of living cells were quantitatively determined by applying MTT test at weekly basis, and cytotoxicity was determined. The potential for adhesion tissue formation was determined by histochemical staining using alcian blue, safranin-o and hematoxylin-eosin, by taking thin sections by freeze sectioning method. Collagen type-1, Anti-podoplanin, Anti-vimentin and Anti-thy-1 antibodies, which are structural proteins found in HDM, were stained by immunohistochemistry, and important matrix components and occurring cellular changes were determined. As a result of these studies, it was determined that the sodium alginate film produced can prevent adhesion and the biomaterial decomposes in a time that may be limited by the healing time of the surgically treated tissues. The histochemical analyzes of the adhesion-like tissue revealed that fibrous tissue was formed on the material, but the amount of tissue formed and the number of cells were very limited. In addition, it was observed that the film structures degraded by 90% on the 21st day, and the adhesion-like fibrous structure remained in free form. The results were also confirmed with SEM analysis images. Within this master's thesis, it was determined that the biopolymeric material produced in cell culture purity could be used in in vivo adhesion experiments and could be commercially brought to the country's economy. It is believed that the created alginate may be used to create cell culture pure biopolymers, which can then be exported for use in the nation's economy.
