Investigation of blood brain barrier development in gestational diabetes mellitus

Abstract (EN)
This study was conducted to examine the development of the blood brain barrier in postnatal gestational diabetes. 18 female mice and 63 offspring obtained were used. Female mice were randomly divided into 3 groups as Control, Sham and Gestational Diabetes. Normal birth periods of mice mated were expected. No application was made to the control group. Intraperitoneal sodium citrate buffer was administered to mice in the sham group on the 5th day of their pregnancy. 40 mg / kg streptozotocin was administered to mice in the Gestational Diabetes group on the 5th day of their pregnancy. Brain tissues were taken from the offspring from the females at P: 0, P: 7 and P: 21 days, with 7 subjects in each group. Serial sections of 5 μm were taken and examined histologically and immunohistochemically. Cell count was made in the prefrontal cortex with a semi-quantitative method developed by our study group. As a result, when Gestational Diabetes groups were compared with Sham and Control groups, it was observed that maternal diabetes significantly decreased the number of cells in the prefrontal cortex. In our study, it was observed that gestational diabetes significantly decreased the expression of these molecules in the immunohistochemistry staining of VE-Kaderin-5 and Klaudin-5, which are tight junction complexes of endothelial cells of the blood brain barrier. Significant decrease was shown in astrocyte marking as a result of Aqp-4 staining. It was observed that maternal diabetes had no effect on pericyte cells. According to these results, we predict that gestational diabetes has negative effects on the development of the blood brain barrier in the newborn and increases permeability.
