Investigation of the effect of cannabinol and 5-fluorouracil on MDA-MB-453 and hela cell lines

Abstract (EN)
This study aims to examine the cytotoxic, morphological and molecular effects of CBN (cannabinol) and 5-FU (5-fluorouracil) on MDA-MB-453 and HeLa cell lines and serves as a preliminary study for the development of cancer drugs in other studies. Cancer is an important public health problem as it is the second cause of death after cardiovascular diseases both in the world and in our country, and it is a difficult disease to combat with its social, material and spiritual aspects, as well as the physical disorders it carries with it. Various treatment methods such as surgery, chemotherapy and radiotherapy are used in cancer treatment. However, these treatments cannot be said to be a complete solution for advanced cancer patients after metastasis occurs. Medication alone is not a solution either. Drug resistance also occurs in patients with drug treatment. Drug resistance in cancer can cause treatment failure. Therefore, it is important to understand the disease and develop treatment methods for it. Recently, research has been conducted on how plants affect cancer cells and it is aimed to find different treatment methods. As a result of the studies carried out, more research needs to be done and unknown properties to be revealed in order to obtain detailed information about plants and their compounds. In this study, we aimed to examine the effects of plants and the use of this plant with drugs on the MDA-MB-453 breast cancer cell line and HeLa cervical cancer cell line. Optimization studies were carried out to determine the effect of drugs on cell lines. When we look at the results of migration and invasion experiments, CBN and 5-FU drugs alone prevented metastasis on cell lines at varying levels, but in combination caused the cells to die. Drugs applied to 3D-developed cell lines caused the 3D structures to disintegrate within 48 hours. According to the molecular docking results of the proteins determined according to molecular docking studies with CBN, the binding energies were determined to be between -5.83 kcal/mol and -98.4 kcal/mol. According to the molecular docking results between our other drug, 5-FU, and proteins, the binding energies were determined to be between -3.4 kcal/mol and -55.7 kcal/mol. In the evaluation made by taking into account the H bonds and binding energy established between the drugs, it was determined that BCL2 protein showed the best cancer activity together with CBN and 5-FU drugs.
