Determination of DNA in Salvia species by capillary gel electrophoresis

Abstract (EN)
In this study, DNA fragments of ten different Salvia (sage) species from Lamiacea family, which have a wide range in Turkey Flora and many of the endemic species, were determined by using agarose gel electrophoresis and capillary gel electrophoresis. For this purpose, the genomic DNAs were firstly isolated from the leaves of Salvia species collected from Eskişehir region. Some parts of genomic DNAs were amplified using OPA-03, OPA-04, OPA-05 and OPA-15 oligonucleotide primers by RAPD method. The DNA fragments were seperated by agarose gel electrophoresis, and these DNA bands were visualized by irradiating with a UV light source. Phylogenetic relation for these Salvia L. species were investigated. The results indicate that the furthest genetically species are S. fruticosa and S. dichroantha, while the most genetically similar species are S. bracteata and S. fruticosa.Dynamic coating method was developed using hydroxyethyl cellulose by capillary gel electrophoresis for DNA separation. An effective separation and reproducible results were obtained for 14 different size of standard DNA fragments by the method. DNA fragments selected from Salvia L. species were determined by capillary gel electrophoresis. The obtained results were also compared with those results by the agarose gel electrophoresis and they were consistent with each other.Keywords: Capillary Gel Electrophoresis, Agarose Gel Electrophoresis, Plant DNA, Salvia,RAPD, Phylogenetic Tree.
