Free radical metabolisims in Carbon Tetrachloride experimental cirrhosis in rat liver; antioxidant effect of Stobadine

Abstract (EN)

In this study, a rat model of CCl4 -induced experimental cirrhosis was used in rats toexamine the level of MDA in order to investigate the oxidative stress and lipid peroxidation.Levels of SOD GSH-Px, CAT enzymes, which prevent free radical injury, were alsomeasured to investigate the effect of stobadinin which is a strong antioxidant.Forty male Wistar-albino rats weighing approximately 250-300 grams were used inthe study. The rats were divided into 4 groups: group 1-control (K), group 2-stobadin (S),group 3- CCl4,(C) and group 4-stobodin+ CCl4 (SC).Cirrhosis was achieved by giving CCl4 intraperitionealy in sequentially increasingdoses. Stobadin treatment was applied orally at a dose of 24.7 mg/kg/day over an 8 weekperiod. All the parameters were measured by using a spectrophotometer.When compared to the control group, while SOD, CAT and GSH-Px enzyme activitiesdecreased statistically significantly (p<0.05), levels of MDA increased statisticallysignificantly (p<0.05).When compared to the control group, while SOD and GSH-Px activities statisticallysignificantly decreased (p<0.05), CAT enzyme activity and levels of MDA increasedstatistically significantly (p<0.05) in the group that was given CCl4+Stobadin.When the group that was only given stobadin was compared to the control group, SODenzyme activity statistically significantly decreased (p<0.05) while CAT enzyme activitystatistically significantly increased (p<0.05). No statistically significant changes wereobserved in GSH-Px enzyme activity and levels of MDA in the same group (p>0.05).When we compared the group that was only given stobadin with CCl4 group, SODGSH-Px and CAT enzyme activities statistically significantly decreased (p<0.05) and thelevel of MDA statistically significantly increased (p<0.05).When we compared the group that was only given stobadin with stobadin+CCl4group, SOD, GSH-Px and enzyme activities statistically significantly decreased (p<0.05) andthe CAT enzyme activity and the level of MDA statistically significantly increased (p<0.05).When we compared the group that was only given CCl4 with stobadin + CCl4 group,SOD enzyme activity and the level of MDA statistically significantly decreased (p<0.05) andCAT enzyme activity statistically significantly increased (p<0.05).In the same group, no statistically significant changes were observed in GSH-Pxenzyme activity (p>0.05).We suggested that GSH-Px enzyme more important antioxidant enzyme in this studycompared with catalase enzymes levels because of GSH-Px activities supressed; but CATactivities increased in group stobadine +CCl4The severe fibrosis was observed in histopathological differences in group CCl4In group stobadine +CCl4; when compared to CCl4 group and control group; there wasa statistically significant improvement was observed althought there wasn?t any compared tocontrol group.
