Abstract (EN)

The importance of shear stress is accepted for endothelial cell layer to perform its barrier function. In this study, in cell culture condition after confluence of bovine endothelial cells on collagen gel, a certain value of shear stress was applied. The aim of this study is examination of new constructed shear stress application model via a hypothesis which claims increase on endothelial permeability monolayer via application of shear stress. This hypothesis is supported with many studies. For this purpose, 5 dyne/ cm2 shear stress was applied on endothelial cell monolayer for 1 hour in flow chamber. Measurement of permeability was performed by using confocal microscopy. In conclusion, it has been observed that diffusion of fluorescent stain labeled dextran (FITC-dextran, Molecular Weight: 77 000 dalton) into collagen gel in cell monolayer which fluid flow was applied for 1 hour, is more deeper than the control group which fluid flow wasn't applied. These results showed that there is accordance with the hypothesis which claims an increase on permeability via application of shear stress and also results showed that new constructed shear stress application model is suitable for endothelial permeability researches.
