Abstract (EN)
Quercetin is a flavonoid whose dose-dependent antinociceptive effect has been demonstrated by different studies. The low water solubility of quercetin, chemical instability and low bioavailability is caused to the limitation of many biological effects. It is aimed to increase bioavailability with a different carrier system to prevent the restriction of the effect profile of quercetin. The aim of this study is to demonstrate the central analgesic effect of quercetin at low doses (3 mg/kg, 5 mg/kg, 10 mg/kg ve 20 mg/kg) with time-dependent acute analgesia tests such as a hot plate and tail immersion tests. In the study, at the same time, quercetin loaded cyclodextrin formulation was created and the analgesic effect observed at low doses and the changes in this effect were compared with pure quercetin doses. According to our research results, it has been shown that the β-cyclodextrin complex increases the solubility in water of quercetin in vitro conditions. The cyclodextrin-quercetin formulation showed a significant analgesic effect compared to control at all applied doses. While pure quercetin did not show a significant effect in both analgesic tests at a dose of 3 mg/kg, the cyclodextrin-quercetin complex showed a significant effect. The cyclodextrin-quercetin complex provided us an easier solubility, an increase in analgesic effect, and ease of application compared to pure quercetin.
