Analgesic effect of kersetin, mechanism of effect and synergistic interaction with different analgesic drugs

Abstract (EN)
It is known that quercetin, which is a bioactive flavonoid, has anticancer, anti-viral, antioxidant and anti-inflammatory effects. In this thesis, it is aimed to investigate the central antinociceptive effect and mechanism of action of quercetin. For this purpose, balb-c female rats which are frequently used to evaluate central nociception in hot plate and tail dipping tests, were randomly distributed to the groups. Quercetin was administered intraperitoneally at doses of 15, 30, 60 and 120 mg/kg. As positive control 30 mg/kg tramadol was used. According to the data obtained, quercetin was evaluated for analgesic effect and a dose of 60 mg / kg was chosen to investigate the mechanism of action. In order to investigate the involvement of the opioidergic system for the evaluation of mechanisms mediating central analgesic activity, non-selective opioid antagonist naloxone (5 mg / kg; ip) were administered 15 minutes before injection of the test substance, 5-HT2 receptor antagonist ketanserin (1 mg / kg; ip), 5-HT3 receptor antagonist ondansetron (1 mg / kg, ip) and to examine the involvement of the adrenergic system alpha-1 receptor antagonist prazosin (1mg / kg, i.p.) and alpha-2 receptor antagonist yohimbine (1 mg / kg, i.p.) were administered 30 min before injection of the test substance. At all administered doses, Quercetin showed significant antinociceptive effect in experimental pain models. According to the results, it was determined that the adrenergic, serotonergic and opioidergic system known to play a role in the modulation of pain plays a role in the antinociceptive effect of quercetin in both hot plate and tail immersion test. It was also observed that the combination of 15 mg / kg quercetin and 15 mg / kg tramadol had a more significant effect than the analgesic effect of the same doses of quercetin and tramadol. As a result; We can say that, quercetin, which has an antinociceptive effect that defined in our study, can be developed alone or in combination with other drugs as an analgesic drug.
