Development of paraoxon based sensors for determination of chemical warfare agents

Abstract (EN)
In this study; Four different methods for detecting chemical warfare agents have been developed. Paraoxon molecule inhibits irreversibly the enzyme acetylcholinesterase. It is preferred on account of resembling nerve agents. As first method of study, quartz crystal microbalance (QCM) based sensor has been developed for determination the paraoxon molecule. QCM's electrode surface is coated with acetylcolinesterase(AChE) enzyme immobilized nanofibers. With the contact of analyte solution to electrode, paraoxon molecules are connected to the AChE enzyme which is on the surface of electrode. With the result of this interaction, the analysis is performed with utilizing the changes occuring in the mass. As second method of study, it is used the nano enzyme system which is synthesized according to method of photosensitive crosslinking with ruthenium based amino acid monomers (ANADOLUCA). With this purpose, AChE enzyme comprising nano enzyme structure was obtained by synthesizing with ANADOLUCA method. The analysis was performed utilizing fluorescence which is changed with the result of interaction between the analyte and synthesized nano enzyme. As third method of study, refractometric interference spectroscopy based sensor has been developed which recognise paraoxon molecule and bind it. With this purpose, carboxymethyldextran coated sensor system is activated and AChE enzyme is binded to this active surface. Changes has occured in the intensity of reflected beam with the result of interaction between analyte and prepared sensor system. The analysis is performed by taking advantage of this change. As last method of study, potentiometric sensor has been developed. Paraoxon imprinted polymer was prepared with this purpose. The prepared polymer has been used as recognation layer in the sensor system. With the result of interaction between analyte and potentiometric sensor, changes has occured in the measured potential. With this changes the analyse is performed.
