Evaluation of apoptotic effects on cancer cell lines clomipramine and chloropyramine

Abstract (EN)

Cancer has attracted the attention of the scientific world due to the fact that in recent years the incidence and mortality rates rapidly increased both in our country and in other countries and it has become a kind of disease which is being investigated for the treatment and tried various methods. Today, cancer is the second leading cause of death after cardiovascular diseases in developed countries. Within the scope of this thesis study, apoptotic and cytotoxic effects of an antidepressant derivative Clomipramine Hydrochloride, an antihistaminic derivative Chloropyrimine Hydrochloride and as a positive control, chemoterapeutik agent anticancer drug Cisplatin were investigated on lung carcinoma cell line (A549) and pancreas cancer cell line (PANC-1). Cytotoxic analyzes of the substances, MTT (3-(4,5-dimethylthiazol-2)-2,5-diphenyl tetrazolium bromide) method, performed on both cell types. Furthermore, antiproliferative effects were investigated in Real Time Cell Analysis System (RTCA MP) and IC50/2 and IC50 values for 24 and 48 hours were determined. Apoptotic effects of Clomipramine Hydrochloride and Chloropyrimine Hydrochloride on A549 and PANC-1 cells researched in detail by using Flow Cytometer and morphological imaging methods. The 24 and 48 hour apoptotic effects of the substances were determined in A549 and PANC-1 cells on the Flow Cytometer using Annexin V / PI, caspase 3 activation, and mitochondrial membrane depolarization (JC-1) methods. Morphological changes of cells were visualized using Confocal Microscopy and Transmission Electron Microscopy. As a result of the study, Clomipramine and Chloropyrimine have reached the conclusion that they may have apoptotic effects on the A549 and PANC-1 cells.
